Single embryoid body formation in a multi-well plate Electron. J. Biotechnol.

Embryoid bodies (EB) formed from murine embryonic stem (ES) cells recapitulate many aspects of a developing embryo. Of specific importance, synchronous differentiation of EB recapitulates organ-specific development and is achieved in culture by formation of uniformly sized EB. The method described here demonstrates a simple and cost-effective way of generating EB from murine ES cells. Single EB are formed in a multi-well plate format and large numbers of EB are generated using a 96-well multi-well plate. Uniform single-sized EB formed in the multi-well are an ideal system for screening compounds and determining differentiation effects. We studied the karyotypes of three hybrid clones of mouse embryonic stem cells and murine splenocytes (two having near diploid and one having near tetraploid chromosome numbers) and the characteristics of their differentiation during the formation of embryoid bodies. The X chromosome originating from embryonic stem cells may be lost in hybrids with a near diploid chromosome number and reprogramming of the "somatic" X may occur. Nuclear transfer embryonic stem cells (ntESCs) show stem cell characteristics such as pluripotency but cause no immunological disorders. Although ntESCs are able to differentiate into somatic cells, the ability of ntESCs to differentiate into primordial germ cells (PGCs) has not been examined. Tumorhead (TH) regulates neural plate cell proliferation during Xenopus early development, and gain or loss of function prevents neural differentiation. TH shuttles between the nuclear and cytoplasmic/cortical cell compartments in embryonic cells. In this study, we show that subcellular distribution of TH is important for its functions. Targeting TH to the cell cortex/membrane potentiates a TH gain of function phenotype and results in neural plate expansion and inhibition of neuronal differentiation. Olivines in main-group pallasites: magma-ocean cumulates or partial melting residues? ArchiMer

Main-group pallasites (MGPs) are meteorites mainly composed of Fe-Ni metal and olivines, the latter being considered as one of the largest sampling of extraterrestrial mantle material available for study on Earth. We analysed the rare earth element (REE) concentrations of olivines from six MGPs to understand better the processes of formation of their parent mantle. All the investigated samples display very low REE abundances, and enrichments in both light REEs and heavy REEs. We interpret the light REE enrichments as a fingerprint of terrestrial contamination. The least contaminated olivines have higher heavy REE enrichments than those inferred for olivines directly crystallised in a magma ocean. The present ultrastructural study on developing and regenerating feathers of chick and zebrafinch describes the ultrastructural changes that occur during the differentiation of barb cells that leads to the formation of the ramus of barbs. Differently from barbule and barb cortical cells that accumulate feather keratin, barb medullary cells undergo to lipid degeneration. Eventually, lipids disappear and medullary cells become empty cavities in the central part of the ramus. Species of the didelphid genera Gracilinanus and Cryptonanus are morphologically and cytogenetically very similar. Several qualitative characters, some of which exhibit intraspecific polymorphisms, have been used to distinguish these genera, but more data are needed to characterize them better. Samples of G. agilis and Cryptonanus spp. from nine localities in central and northeastern Brazil were analyzed. Electrophoretic protein pattern and acid phosphatase activity in the midgut extracts of Apis mellifera L. (Hymenoptera: Apidae) during metamorphosis Neotropical Entomology

The electrophoretic pattern of proteins and the activity of acid phosphatase were described in the midgut extracts during the post-embryonic stages of Apis mellifera L. in order to stablish a correlation with the metamorphosis events. The results show the greatest variation in proteic electrophoretical bands during prepupal stage and the smallest variation during the brown eyed pupae stage. The acid phosphatase activity was higher during the larval last instar and lower in white eyed pupae. The greatest variety of bands during the prepupal stage coincides with the histolysis of the larval midgut epithelium and the lowest variability in the brown eyed pupa coincides with the end of the midgut differentiation. This paper analyzes the effect of competition on product demonstration decisions. Pre-purchase product demonstration enables marketers to differentiate products that are ex-post differentiated but are judged according to perceived fit, rather than actual fit, due to pre-purchase consumer uncertainty. Imbalanced competition accompanied by fit uncertainty motivates the follower to offer demonstrations to avoid a price war. This paper explores the conditions that lead the leader to retaliate. Due to a high level of product substitution in the food market, it often proves difficult to strike a balance between supply and demand. This is especially true in sectors with a such high level of competition and product differentiation as the wine producing sector. Faced with the difficulty of differentiating the product in terms of its technical characteristics, quality and price, therefore, a useful alternative is to explore what consumers perceive to be its "emotional benefits", since these have been shown to have a decisive effect on consumer purchasing decisions. This paper shows that retailers may choose to offer products differentiated in quality to consumers, not to relax downstream competition, but to improve their buyer power in the negotiation with their supplier. We consider a simple vertical industry where two producers sell products differentiated in quality to two retailers who operate in separated markets. In the game, first retailers choose which product to carry, then each retailer and her chosen producer bargain over the terms of a two-part tariff contract and retailers finally choose the quantities. Coffee Differentiation: Demand Analysis at Retail Level in the US Market AgEcon

Scanned data was used to estimate US coffee demand using an AIDS model. The estimated elasticities have the expected signs and magnitude. Differentiated coffees are complements for regular and unclassified while regular and unclassified coffees are substitutes. These results could be useful in designing marketing strategies by coffee suppliers.

The paper analyzes competition among supermarkets in Brazil. In contrast to part of the economic literature which suggests that the fast growth of big supermarket chains would destroy independent, medium and small supermarkets, the paper argues that big supermarket chains can coexist with different formats of independent food retailing. As a result, competition in food retail is complex and cannot be described as a simple Darwinian process of market concentration. The analysis is divided in two parts. A semi-nested reverse transcription-polymerase chain reaction (Semi-N-RT-PCR) was developed and used to detect the S glycoprotein gene of infectious bronchitis virus (IBV) strains and to discriminate H120 vaccine strain from other strains. Viral RNA was extracted from the allantoic fluid of chicken embryos and from tissues of chickens experimentally infected with different strains of IBV. The objective of this study was to determine consumers understanding of beef traceability, identifying how consumers value this meat and traceability elements to be presented on retail shelves. The method used in this study was a survey through the internet applying the Sphinx software. The sample consisted of 417 consumers, mostly living in Porto Alegre, Brazil. Consumers are aware of certified beef, consider it important, but this is not a demand. As to traced beef, most consumers (62.4%) are in favor of mandatory traceability of beef cattle in Brazil, but 86.6% disagree with the destination of traced beef only to the foreign market.
